You also want to make sure that you are properly attired and that you get another person to help you. This is really a two person job. Both of you should have on heavy duty gloves, clothes that cover all your body so no skin is exposed, and a mask would be a good idea as well. Just to be safe.

Your objective is to scare the squirrel into the bag. Then when it runs into it the other person closes it up and you take it outside and get rid of it. Use some kind of a stick or a shovel to nudge the squirrel toward the bag. The most important thing is to keep it inside the enclosed area.
